The limiting step is getting DNA from outside to inside the cell. The more DNA there is, the more likely some molecules will slip through the membrane.

    The efficiency does not necessarily increase as you increase the amount of DNA added. Now, typically the amount of DNA inside the cell will increase as the amount outside increases, but the efficiency does not. Transfection efficiency is usually more dependent on the vector and the method.
